Recent research indicates that GLP-1 drugs, such as semaglutide—the active ingredient in medications like Ozempic and Wegovy—may play a role in reducing the risk of dementia. A meta-analysis involving 26 clinical trials and over 160,000 participants has provided evidence suggesting that these drugs can significantly lower the chances of developing dementia or cognitive impairment when compared to placebo treatments.

The study, led by Catriona Reddin at the University of Galway, focused on individuals with type 2 diabetes who had not been diagnosed with dementia. The findings build on previous observational studies that hinted at a link between GLP-1 drugs and reduced dementia risk. Unlike earlier research, which was primarily based on health records, this analysis utilized randomized trials, thereby strengthening the evidence for the protective effects of these medications.

While diabetes is known to increase the risk of dementia, the protective mechanism of GLP-1 drugs appears to extend beyond mere blood sugar control. Research suggests that these drugs may reduce neuroinflammation, a factor increasingly recognized as contributing to dementia. Additionally, GLP-1 drugs may mitigate cardiovascular issues, such as plaque build-up and high blood pressure, which can also impact cognitive health.

Experts like Ziyad Al-Aly from Washington University acknowledge that while these findings are promising, they do not yet warrant prescribing GLP-1 drugs solely for dementia prevention. However, physicians may consider these medications for diabetics who are at heightened risk of cognitive decline.

Reddin emphasizes the need for larger, long-term trials to further investigate the effects of glucose-lowering therapies on dementia. Two studies exploring semaglutide’s potential in treating early Alzheimer’s disease are expected to conclude later this year.
